چکیده

Propranolol, a widely used beta-blocker, inhibits the hydrolysis reaction of enzyme cholinesterase. Measurements of the difference in rate of hydrolysis rate between uninhibited and inhibited reactions allow the development of a kinetic method for its determination. Both systems, enzyme–substrate–chromogen and enzyme–substrate–chromogen–inhibitor, were characterized through biochemical kinetic parameters (KM, 0.326–0.330 mmol/L; Vmax , 40.0–43.0 μmol/Lmin). The inhibition type was recognized as competitive and the inhibition constant, Ki, was determined to be 22.60 μmol/L. The detection and quantification limits were calculated as 0.004 and 0.0136 μmol/L, respectively. Accuracy and precision of proposed methods were tested. The proposed method showed good sensitivity, selectivity, simplicity and rapidity, thus it is convenient for clinical applications.

Titrimetric and Spectrophotometric Methods for the Determination of β-blockers in Pharmaceutical Dosage Form

Three new methods using titrimetric and spectrophotometry are described for the determination of Propranolol Hydrochloride (PPH) and Atenolol (AN) with potassium bromate as the oxidimetric reagent and acid dyes, Methyl orange and Indigo carmine. In direct titrimetric (method A), the drug is titrated directly with bromate in acid medium and in the presence of excess of bromide using methyl orang...

Kinetic-Spectrophotometric Determination of Thiocyanate Based on Its Inhibitory Effect on the Oxidation of Pyrogallol Red by Bromate

A kinetic spectrophotometric method for rapid measurement of thiocyanate in serum, urine and sewage water with no need for removed of interfering substances in described. The method is based on the inhibitory effect of thiocyanate on the oxidation of pyrogallol red by bromated and nitrite which is monitored at 467 nm. The variables affecting the rate of the reaction were investigated and th...

KINETIC SPECTROPHOTOMETRIC DETERMINATION OF MERCURY IN DIFFERENT MATERIALS BASED ON ITS INHIBITORY EFFECT ON THE BRILLIANT GREEN-SULFITE REACTION

A new kinetic method for the determination of Hg(II) based on its inhibitory effect on the Brilliant green-sulfite reaction at pH 6.5 is proposed. The reaction is followed spectrophotometrically at 615 nm by measuring the change in absorbance at the fixed time of 3.5 min after the initiation of the reaction. Both the influence of the reaction variables and the interference of many ions have...

Conductometric determination of propranolol hydrochloride in pharmaceuticals

______________________________________________________________________________ In this paper the conductometric titration of propranolol hydrochloride in pharmaceutical formulations using silver nitrate as titrant is proposed. The method was based on the formation of an insoluble salt (AgCl(s)) between the chloride of propranolol hydrochloride molecule and Ag(I) ions of the titrant AgNO3. The e...
